Sometimes, Mothron slows down, lands, and lay an egg, while being a static target for a couple of seconds. Using a weapon that hit multiple times (the death scythe in your screeshot, for example) at this moment is nice, as it will damage Mothron a lot and the egg that will spawn under it.

When should I kill Duke Fishron?
Take him on after Plantera, or maybe after the Moon events. Even end-game players sometimes have trouble with the Duke. I took him out after laying down a skybridge made of Asphault blocks, and even then I had a chance of de-spawning it if I go too fast.
